NMRI اثر رتینوئیک اسید بر تکوین بیضهای موش کوچک آزمایشگاهی نژاد

<p dir="RTL"><strong>زمینه و هدف: </strong>رتینوئیدها، مولکولهای مهمی هستند که فرآیندهای اساسی نمو را در تمام مهرهداران تنظیم میکنند. هدف مطالعه حاضر، بررسی اثر رتینوئیک اسید بر فرآیند نمو بیضه است.</p>
<p dir="RTL"><strong>مواد و روشها: </strong>یک گروه از موشهای تازه به دنیا آمده نژاد <span dir="LTR">NMRI</span> تحت تزریق داخل صفاقی رتینوئیک اسید به میزان <span dir="LTR">mg/Kg.b.w</span> 25 قرار گرفتند و گروه دیگر بدون تزریق بودند. پس از 21 روز حیوانات نر جدا و در 60 روزگی کشته شدند و بیضهها از بدن آنها خارج گردید. از هر گروه، هفت سر موش از لحاظ ویژگی ظاهری بیضه مورد مشاهده قرار گرفتند. در بررسی میکروسکوپ نوری، شمارش سلولی اپی تلیوم لولههای منی ساز انجام شد و قطر لولههای منی ساز اندازهگیری شد و با آزمون آماری <span dir="LTR">T-test</span> بین گروههای کنترل و آزمایش مقایسه صورت گرفت.</p>
<p dir="RTL"><strong>نتایج:</strong> وزن بیضه در مقایسه با گروه کنترل تغییر معنیداری نداشت. در اپی تلیوم زایای بیضه، ضخامت اپی تلیوم و تعداد سلولهای اسپرماتوگونی، اسپرماتیدهای گرد و طویل شده در گروه آزمایش نسبت به گروه کنترل کاهش معنیداری نشان داد (<span dir="LTR">P<0.05</span>).</p>
<p dir="RTL"><strong>نتیجهگیری: </strong>بهکارگیری رتینوئیک اسید پس از تولد، بر نمو بیضه و اپی تلیوم لولههای منی ساز آن، مؤثر است. حتی دوز ناچیز آن بر تقسیمات سلولی اپی تلیوم منی ساز اثر مخربی دارد بهطوریکه تعداد سلولهای زایا تحت تأثیر آن کاهش مییابد؛ لذا در استفاده از ویتامین <span dir="LTR">A</span> و مشتقات سنتتیک آن مانند رتینوئیک اسید برای مادران باردار و شیرده باید احتیاط لازم صورت گیرد.</p>
<p style="margin-left:-7.15pt;"><strong><em>Background & Objectives:</em></strong> Retinoids are important molecules that regulate crucial processes of development in all vertebrates. In this article, we study the effect of retinoic acid on testis development.</p>
<p style="margin-left:-7.15pt;"><strong><em>Materials & Methods: </em></strong>A group of newborn NMRI mice was chosen to receive intraperitoneally injections of 25 mg/Kg.b.w of retinoic acid and the control group had no injection. After 21 days, the male animals were isolated and sacrificed in 60 days postpartum, and testes were removed from their bodies. Apparent characteristics of the testes of seven mice from each group were observed. In the light microscopic study, these epithelial cells of the tubules were counted and the diameter of seminiferous tubules were measured with statistical T-test analysis and were compared with control group.</p>
<p style="margin-left:-7.15pt;"><strong><em>Results:</em></strong> The findings showed that the weight of the testes in animals that were affected by retinoic acid did not change significantly compared to control group. In germ epithelium of testis, the thickness of the epithelium and the number of the spermatogonia, round and elongated spermatids has decreased significantly compared to the control group (P<0.05).</p>
<p style="margin-left:-7.15pt;"><strong><em>Conclusion:</em></strong> According to the findings of this article, using the retinoic acid after birth has influence on testis development and its seminiferous tubules epithelium. Retinoic acid has an adverse effect on the cell divisions in seminiferous epithelium, therefore the number of affected germ cells decrease. Therefore, the use of vitamin A and its synthetic derivatives like retinoic acid for pregnant women should be done with caution.</p>
